Antibodies / Biomarkers: Recombinant Antibodies in Pathology

The use of antibodies in immunohistochemistry has had a major impact in pathology in the last couple of decades. During this time, the role of immunohistochemistry has evolved from an ancillary technique to a major diagnostic and prognostic tool (e.g. HER2 neu in breast carcinoma). The panel of antibodies available to pathologists has expanded rapidly, due to the development of efficient methodologies for antibody production (e.g. hybridoma). In addition, recombinant antibody technology has become a potentially promising tool in diagnostic pathology.

Recombinant antibodies are produced by a method known as phage display, whereby antibody fragments (e.g. single variable Fv fragments) containing specific binding sites of the parent antibody are cloned and displayed on bacteriophages secreted by infected bacteria (usually Escherichia coli). In this way, antibody “libraries” can be constructed that facilitate isolation of antibody fragments to particular antigens. Advantages of recombinant technology include increased consistency across antibody lots, production of antibodies to highly toxic antigens, and modification of antibody fragments to enhance specific characteristics such as binding affinity. As a result of phage display techniques, “customized” antibodies are becoming increasingly available from biotech companies (e.g. AxioMx, Branford, CT) and will likely complement the trend towards personalized medicine in the coming years.
