Grossing Equipment: Tissue Embedding Cassettes

Tissue embedding cassettes are one of the essential tools of the anatomic pathology laboratory. In addition to the standard slotted cassette designed for 3 mm thick tissue sections, various other cassette designs are available with specific features to optimize fixation and orientation of different tissue specimens. Some examples include 1 mm square mesh and microcompartment cassettes for biopsies, and larger cassettes that can accommodate up to 10 mm thick tissue sections (e.g. for prostate or ophthalmic tissue). Many laboratories employ tissue cassette labelers that apply a unique barcode and case number to each cassette, thus saving time, reducing errors, and facilitating block identification and retrieval.
With the advent of automated tissue embedding, new innovative products have been developed with the goal of increasing and streamlining workflow while reducing tissue orientation errors and loss. One example is the Tissue-Tek® Paraform® Sectionable Cassette System by Sakura. This cassette system is comprised of an outer cassette frame containing an inner cassette made from Paraform®, a material amenable to sectioning like paraffin wax. Once the tissue is placed inside the Paraform® cassette, tissue orientation is preserved throughout the process of embedding and sectioning. This system may be especially useful in small biopsies where orientation is critical for accurate diagnosis (e.g. certain skin or gastrointestinal biopsies).
