Physiology

● Sequential processing of the beta-amyloid precursor protein (APP) by beta and gamma secretase generates the amyloid beta-peptide (A beta), which likely has a causative role in Alzheimer's disease (AD)
● A beta 42 is a 42 amino acid variant of A beta, believed to be pathogenic
● A beta 42 may trigger the amyloid cascade, leading to formation of plaques
● A beta 42 is usually a minor species produced by APP cleavage, but its production is enhanced by familial Alzheimer Disease mutations in presenilin 1 (PS1) and PS2, as well as by some familial Alzheimer Disease mutations in APP (J Biol Chem 2011 Feb 25 [Epub ahead of print])
